How ‘777’ Oil works against Psoriasis? A mechanistic approach

Psoriasis is a polygenic, chronic relapsing inflammatory immune-mediated disease based on interplay between antigen-presenting dendritic cells, T cells and keratinocytes creating the typical signature skin lesions, which are sharply demarcated reddish and scaly plaques. It devastates patients and, all too often, to doctors who treat them. Advances in the understanding of the immune basis of psoriasis have led to the development of therapies targeted at immune/inflammatory pathways leading to psoriasis.

In this context it has become imperative to search for alternatives preferably from the Herbal and natural world.

‘777’ oil, a sidhha formulation in which the main ingredient is Wrightia tinctoria (Vetpalai leaves) a folklore claim was found to be highly beneficial in the treatment of psoriasis. It is a single herbal formulation prepared by lipid extraction procedure. This drug is found to be effective in the management of Psoriasis. This formulation has already been clinically evaluated several thousand of patients benefited across India and many parts of the world and has been proven to be very effective for long-term usage. It is used for all the cases as external application and the patients were advised to have a thorough bath in Luke warm water using green gram and Bengal gram flour.  After drying the body with clean towel they were instructed to apply the oil generously over the lesions and advised to expose themselves in the early morning sun light for 15 to 30 minutes.  The oil is left to remain on the body till the next day.  This process is repeated daily.

MECHANISM OF ACTION OF ‘777’ Oil

The pre-clinical and clinical trials conducted over the years revealed that the application of ‘777’ Oil retards the cell division and inflammatory responses. It also has immunomodulatory properties.

Reduction in Cell Multiplication

‘777’ oil significantly reduces the cell multiplication mainly in late metaphase and early anaphase event of mitosis. In vitro study of the effect of ‘777’ Oil on cultured cells was carried out using fast growing cell line SKV 14. Significant reduction in cell multiplication was observed. The study was also confirmed in Onion root tip mitosis assay. The rate of cell multiplication retardation is calculated as 40-60%. This property of ‘777’ oil was re-confirmed in Guinea pig. Hyperkeratosis was induced in the flank region of the Guinea pig using a mutagen -Ethidium Bromide. 777 Oil was applied parelelly to study its effect on hyperkeratisos. It was recorded that 777 Oil arrests the level of cell multiplication to 58-63%. The isolated cells were further stained with fluorescent dye and found that spindle and microtubule formation has occurred in the treated cells.

Antiinfllammatory property

For evaluation of anti-inflammatory activity, cytokine ELISA was done using the samples to study the co-stimulatory molecule profile in the psoriatic condition and the effect of 777 Oil on them. IL80 and Lymphokine 41 were found to have blocked during 777 Oil applications. Receptor binding mechanism would have resulted in this phenomenon. The effect on 777 Oil on the IL 40 may answer for its efficacy on the inflammatory changes of psoriasis.

Immunomodulatory activity

777 Oil was studied for its immunomodulatory property both by in vitro and in vivo techniques. The phagocytes were isolated from the peritoneal cavity of Balp C mice after injecting with thyoglycolate. The phagocytes was adhered on ELISA plate and pre-treated with different dilutions of 777 Oil for 20 minutes. Later the cells were infected with opsonized fungal cells of Candida albicans. The level of phagocytosis and phagocytic killing (phagocytic Index, PI) were calculated. Similarly, 777 Oil was administered orally to mice after injecting with SRBC (Sheep Red Blood Cells) and the level of antibody production was estimated by haemaglutination. Result confirmed that ‘777’ oil nicely modulated the immune response.

Clinical study

The efficacy of 777 Oil was confirmed on 300 cases taken up for trials in the out patient department of Central Research Institute, Siddha at Chennai, India.

Conclusion

Do we still have more to do? We still haven’t found the potency and exact mechanism of all Medicinal Herbs/Formulations used in psoriatic treatment. The search of this mechanism will undoubtedly speed up over the next decade. If it becomes possible then we can expect a way for managing psoriasis that is entirely harmless, very convenient, and entirely safe. Hopefully, with the discovery of mechanisms and scientific validation of natural herbs, we will gain insights into new outcomes for more effective and safer therapies.
